|Job Description :||We are seeking a driven and talented candidate to join our Analytical and Formulation Development group in La Jolla, CA. In this role, the Analytical Development Scientist I will focus on the development and execution of LC-UV assays for large molecules (e.g. mAbs, antibody fragments, and cytokines). As an independent contributor, s/he will collaborate with other functional groups in support of platform development, strain engineering, upstream, and downstream process development based on defined project goals and within established timelines. Key Responsibilities: The successful candidate will be expected to: Apply solid protein analytical scientific knowledge and practical experience to develop HPLC/UPLC separation methods for the characterization of large (e.g. antibodies, fragments, PEGylated proteins) molecules, including by SEC, IEX, RP, HILIC, and HIC, in conjunction with detection by UV and mass spectrometry. Develop and perform LC-UV and/or LC-MS assays to determine the purity and abundance of protein/fragments and small molecules, in support of early-stage Synthorx research programs and throughout the product development cycle. Utilize multiple HPLC instruments for method development simultaneously to shorten the cycle of learning, taking advantage of multiple modes of separation to interrogate & characterize novel molecules (proteins) that emerge from the Synthorx platform in a timely manner. Perform daily & weekly operator maintenance on HPLC instruments used in support of method development and routine sample analysis, and perform troubleshooting with vendor technical support as needed. Manage priorities, plan experiments, conduct analysis, and prepare research reports to support timely turnaround to project teams. Write and review protocols, and enter results into an electronic lab notebook (ELN). Compile and present experimental results in team, project, and inter-site meetings as needed. Basic Qualifications: Required qualifications include: Ph.D. in Biochemistry, Analytical Chemistry or a related discipline with 1+ years in post-doc/industry; Master’s degree with 5+ years, or Bachelor’s degree with 7+ years of relevant full-time work experience in the pharma/biotech industry. Prior hands-on experience with sample extraction for proteins/peptides from biological samples, such as bacterial or mammalian cell pellets, tissue culture media, or serum samples is required. Proficiency in designing and execution of HPLC-based characterization of large molecules in at least one mode is required: IEX, SEC, RP, HILIC, HIC. Proficiency with the use of chromatography data systems for acquisition method setup, data processing, and data processing (Chromeleon, OpenLab, or Empower) is required. Previous experience in performing daily/weekly operator maintenance of HPLC/UPLC systems is required. Prior experience in presenting scientific work to a scientific audience is required. Ability to adhere to good documentation standards and maintain detailed records of all work performed is required. Experience in LC-MS analysis of intact proteins or peptides on QTOF, QqQ, QDa, and/or Orbitrap will be beneficial. Experience in CE for the separation of proteins will be beneficial.
